P311在肺泡发生中的作用初探

摘    要:P311是一个由肺泡形成相关阶段相对于肺泡成熟期差减杂交库中获得的上游调节基因,为探讨其在肺泡发生过程中的可能作用,本以真实时间PCR(realtimePCR)和免疫组织化学方法确定了P311在肺器官发育过程中的表达特点,结果显示P311的表达无论在时间上还是在组织细胞分布上都与肺泡的发生密切相关.同时胎肺细胞植模型中,反义寡核苷酸阻断P311表达的研究亦显示P311在肺泡分隔膜形成过程中的重要性.

Abstract:P311 is an upregulating gene of alveolar morphogenesis, which was obtained from the suppression subtractive hybridization library in which cDNA encoding genes differentially expressed at the period of alveolar formation. Real time PCR was performed to determine the expression profiles of P311 in the different stages of lung development. It showed that P311 was expressed in the whole process of lung development, whereas the peak level only appeared in the stage of alveolization, then dropped to the bottom line in the adult lungs. Immunohistochemical staining exhibited that P311 protein localized on the edges of saccular or alveolar walls directly exposed to the airspaces, and also on the septa. Following treated with antisense-P311, the dissociated fetal lun cells failed to develop to the the precursors of alveoli in the renal capsule graft model. Taken together, these studies suggest the P311 could be involved in regulation alveolar morphogenesis, especially in septation.
