茶多酚对Hep-G2细胞活性及形态的影响

摘    要:大量研究表明,长期饮茶尤其绿茶者,可大大降低各种癌变的发病率,而且对多种体内外肿瘤具有直接杀伤作用.作者通过采用不同浓度的茶多酚作用于体外培养的肝癌细胞株Hep-G2细胞,初步探讨茶多酚对Hep-G2细胞的凋亡作用.采用MTT法(噻唑蓝比色法)和AO-PI(吖啶橙-碘化丙啶)双重荧光染色法研究了茶多酚诱导Hep-G2细胞株的凋亡作用.MTT法研究结果表明,在一定的浓度范围内,茶多酚对Hep-G2细胞有明显的抑制作用,且存在良好的剂量效应关系,随着茶多酚浓度的升高,其对Hep-G2细胞的抑制率也升高.Hep-G2细胞经茶多酚作用后,细胞形态发生明显的变化,可见凋亡小体,荧光强度增强,细胞呈现凋亡现象.茶多酚在体外对Hep-G2细胞的增殖具有抑制作用,诱导其发生凋亡.研究结果为茶多酚的进一步开发应用奠定理论基础.
