低蛋白和高蛋白饲料对大鼠骨代谢的影响

摘    要:应用骨组织形态计量学的方法观察并比较低蛋白和高蛋白饲料对大鼠骨骼的影响。将 3月龄雄性Sprague Dawley大鼠 18只随机分成 3组 :对照组control(Pr2 0 % )、低蛋白组LPD(Pr8% )、高蛋白组HPD(Pr2 3% )。饲喂按AOAC方法合成的提纯饲料 90d后 ,取左侧胫骨近心端不脱钙骨包埋后 ,进行骨组织形态计量学分析。测得结果为 :与对照组相比 ,LPD骨小梁面积百分率 (%Tb .Ar)减少 5 7% (P <0 .0 1) ,骨量明显减少 ,骨小梁间距 (Tb .Sp)增大185 % (P <0 .0 5 ) ,反映骨形成的指标骨形成率 (BFR TV)明显降低 (P <0 .0 5 ) ,荧光周长百分数 (%L .Pm)、矿化沉积率 (MAR)有降低的趋势 ;HPD亦使大鼠 %Tb .Ar减少 40 % (P <0 .0 5 ) ,呈骨低转化的趋势 ;LPD与HPD相比 ,各项指标都有所增强。由上可知高蛋白和低蛋白饲料都使大鼠骨量明显减少 ,骨结构变差 ,骨强度下降。其中低蛋白饲料对骨形成抑制大于高蛋白饲料。

Abstract:To study the effect of low or high protein diet on bone metabolism in male rats by bone histomorphometry. Eighteen 3-month-old Sprague-Dawley male rats were randomly divided into three groups:control group(pr20%)、low protein(pr8%) and high protein group(pr23%).Feeding purified diet for 90 days,all rats were killed.The poximal tibia(PTM) was harvested for histomorphometic analysis. LPD group had lost 57%( P <0.01)of bone cancellous compared with control group.Dynamic date in LPD group showed bone formation indices such as percent labeled perimeter(L.Pm%),mineral apposition rate(MAR) and bone formation rate(BFR/BV) increased without significance;HPD group had also lost 40%( P <0.05) of cancellous bone compared with control group, representing decreased bone turnover.Comparing with HPD group, indicates of LPD group all increased .The results indicated bone cancellous loss evidently、bone frame was weak、bone intension decreased in low protein dietary rats,and high protein dietory rats is also.But LPD restrain bone formation more strongly than HPD.
