99mTcN-GLA的制备及其生物分布研究

摘    要:将99mTcO-4与浓HCl和NaN3在沸水浴的条件下制备99mTcNCl-4中间体,然后与配体葡萄糖二酸(GLA)发生交换反应得到放化纯度大于90%的99mTcN-GLA配合物.小鼠体内生物分布结果显示: 99mTcN-GLA与99mTc-GLA有明显不同的生物分布,特别是99mTcN-GLA的肺摄取及滞留均较好,有望成为一种新型的肺显像剂.

Abstract:99mTcN-GLA (GLA: glucaric acid) is prepared through a ligand-exchange reaction. The two-step procedure involves the initial reaction of 99mTcO-4 with sodium azide(NaN3) and concentrated hydrochloric acid at 100℃ to form the intermediate(99mTcNCl-4), followed by the addition of the ligand GLA. The radiochemical purity (RCP) of the product is over 90% by TLC. The biodistribution results in mice indicate that the presence of 99mTc nitrido group significantly alters the biological properties of the 99mTc-complex formed using the same ligand. 99mTcN-GLA has good lung uptake and retention, suggesting that it will be a potential lung imaging agent.
