银杏叶中总黄酮的提取和测定

摘    要:本试验比较了有机溶剂浸提法和超声波法提取总黄酮的效果,选择提取率较高的超声波法来提取。对料液比、超声时间和乙醇体积浓度对银杏中总黄酮提取效果的影响进行了单因子试验,并采用正交试验法找出最佳提取条件为料液比1g∶30 mL、超声时间30min、乙醇浓度70%。采用紫外可见分光法检测黄酮类化合物的含量,不加显色剂,直接以258 nm作为最大吸收波长。结果表明,银杏中总黄酮的平均含量稳定在2.77%,精密度的RSD为0.302%,该方法的平均回收率为100.3%。本试验方法样品处理简单,准确度高,精密度好,适合于银杏叶中总黄酮的提取和测定。
