摘    要:以针状CaCO3为无机模板,制作成新型固定化材料管状空心SiO2载体,用来固定葡萄糖氧化酶(GOD).研究了pH值,温度和载体与GOD的配比等对固定化酶活的影响。所得的最佳工艺条件为:最佳载体和游离酶配比为0.6g/mL;固定化酶的最适pH值为5.2;最佳反应温度为32℃.和游离酶相比无论操作性还是稳定性都有提高。

Abstract:Porous hollow silica nanotubes (PHSNTs) have been synthesized via a sol-gel route using nano-sized needle-like CaCO3 as the inorganic template and employed as a support for immobilization of Glucose Oxidase enzyme (GOD) biocatalyst. The effect of varying factors such as phosphate buffer pH, immobilization temperature and ratio of carrier to free GOD (g/mL) on the activity of immobilized GOD are discussed. The optimum conditions were found to be as follows: ratio of carrier to free GOD (g/mL) 0.6g/mL, phosphate buffer pH 5.2, immobilization temperature 28℃. The results show that under optimized conditions the efficacy and stability of GOD are improved relative to the free enzyme. 
