西瓜NPR1抗病基因的遗传转化

摘    要:比较了不同预培养时间、侵染时间、共培养时间等因素对农杆菌转化西瓜的影响。西瓜遗传转化的适宜条件:3 d苗龄的子叶,预培养1 d,农杆菌侵染10~15 m in,共培养3 d;不定芽与根的潮霉素(Hyg)筛选浓度分别为20 m g.L-1和6 m g.L-1。对20株西瓜转化苗进行DNA提取,PCR检测,其中5株呈阳性,初步证明目的基因已经整合到西瓜基因组中。

Abstract:The Agrobacterium mediated transformation of Citrullus vulgaris was studied.Effects of different time interval of preculture,bacterial infection and co-cultivation on the transgenic efficiency were investigated.The results showed that preculture three days cotyledon for 1 day was in the best adptive condition,bacterial infection was 10~15 min,and co-cultivation for three days.The adventitious bud and root of watermelon were sensitive to hygromycin,20 mg/L and 6 mg/L was the suitable screening concentration.Transforming plantlets were detected by PCR,five transformants had got from 20 plantlets.The results indicated primarily that the target gene was inserted into watermelon genome.
