摘    要:利用加替沙星自组装环荧光显微成像技术,建立一种灵敏度高、操作简便的加替沙星固相检测方法.在pH 3.60的HAc-NaAc缓冲溶液和聚乙烯醇-124存在下,加替沙星能够在二氯二甲基硅烷(DMCS)处理过的载玻片上形成自组装环(Self-ordered ring,SOR),环直径为2.45 mm,环线宽为38.7μm.当点样体积为0.30μL时,线性范围6.74×10-13~1.83×10-11mol.ring-1,方法的检出限(3σ)为6.74×10-14mol.ring-1(2.2×10-7mol.L-1).应用于内蒙古乳都品牌乳、加替沙星胶囊和健康志愿者服药后尿液等样品中加替沙星的分析检测,加标回收率为96.0%~106.6%,RSD小于3.3%.

Abstract:A highly sensitive and simple solid-state method was introduced for the determination of gatifloxacin based on the capillary effect of solvent on a solid support with fluorescence microscopic technique.In the pH 3.60 HAc-NaAc buffer solution and Poly(vinyl alcohol)-124 (PVA-124), a typical fluorescent SOR of gatifloxacin can be obtained on dimethyl dichlorosilane(DMCS) pretreated hydrophobic glass slide with outer diameter less than 2.45mm and ring belt width less than 38.7μm.Linear response is observed in the range of 6.74×10~(-13)~1.83×10~(-11)mol·ring~(-1)and the detection limit (3σ) reaches 6.74×10~(-14) mol·ring~(-1)(2.2×10~(-7) mol·L~(-1)) when 0.30 μL droplet is spotted.The assay was successfully applied into the determination of gatifloxacin in brand milks of Inner Mongolia,gatifloxacin capsules and human urine samples of healthy volunteers after dosing with the recovery of 96.0%~106.6% and RSD lower than 3.3%.
