双氢青蒿素醚化法制备蒿甲醚的工艺优化

摘    要:采用正交试验法和HPLC分析技术,对蒿甲醚的制备工艺进行优选,以蒿甲醚收率和含量为考察指标,对影响双氢青蒿素醚化的因素进行研究,其醚化的最佳条件为A2B3C4D1,即甲醇与二氯甲烷的体积比为1∶2;双氢青蒿素与三氟乙酸的量之比为10∶1;反应温度为40℃;反应时间为1.5h。该工艺稳定可行。

Abstract:The preparation process is optimized by using an orthogonal design and HPLC. The extraction rate and content of Artemether have been taken for the observed indexes. The best etherificafion condition for dihydroArtemisinine is A2B3C4D1. The ratio of methanol volume over dichloromethane volume is 1 : 2, the rate of accelerating agent dosage 10: 1, the reaction temperature 40℃ and the reaction time 1.5 hours. When meeting all the conditions, the optimal preparation process was feasible.
