MTT比色法在检测重组人GM—CSF生物活性中的应用

摘    要:探索了MTT比色法的一系列实验条件,并利用GM-CSF依赖细胞株TF-1,测定了家蚕基因工程生产的重组人GM-CSF(granulocyte-macrophagecolony-stimulatingfactor)的生物活性,并与半固体培养集落刺激法进行了比较。

Abstract:The cleavage of the tetrazolium salt MTT into a blue coloured product (formazan) by the mitochondrial enzyme succinate dehydro genase is potentially very useful for assaying cell survival and proliferation.The conversion takes place only in living cells and the amount of formazan produced is proportional to the number of cells present. We have established the method that is optimal to the biological activities assay of the rhGM CSF using the TF 1 cell line.Meanwhile,the result is compared to that obtained from colony forming methods.
