摘    要:裂殖壶菌Schizochytrium sp.FJU-512是高产二十二碳六烯酸(DHA)的一株海洋真菌,其细胞内油脂占细胞干质量的18.3%.细胞油脂的总脂肪酸组成为45.7%DHA,12.5%DPA和41.8%棕榈酸.总油脂由26.4%极性脂和73.6%中性脂组成.薄层色谱、高效液相色谱及质谱分析表明该菌株还能合成强抗氧化剂虾青素.采用均匀设计获得高产DHA的最优培养基(g/L):葡萄糖109.7,蛋白胨19.2,酵母膏20.0,海水晶10.4,DHA产量高达2.10 g/L.

Abstract:Schizochytrium sp.FJU-512 was a marine fungus producing a large amount of docosahexaenoic acid(DHA).The content of the total lipid in its dry cell was up to 18.3%.The profile of fatty acids in the total lipid consisted of 45.7% DHA,12.5% docosapentaenoic acid(DPA) and 41.8% palmitic acid.The total lipid contained 26.4% polar lipid and 73.6% neutral lipid.HPLC and MS analysis showed the strain could also produce astaxanthin,which could protect the cell oil against oxidization.The optimum medium for DHA production were as following: glucose(110.0 g/L) as carbon source,peptone(5.0 g/L) together with yeast extract(18.0 g/L) as nitrogen source;salt 12.5 g/L.The highest DHA-production reached 2.1 g/L at the optimize medium.
