中华猕猴桃蛋白酶催化功能基团的研究

摘    要:从野生中华猕猴桃分离提纯得SDS电泳单一纯酶制剂,酶蛋白的总巯基数为7,其中游离巯基数为1.3×10~(-5)MpCMB使酶活力丧失100%,酶修饰失活呈一级反应,表明酶分子的唯一游离巯基为该酶催化活性的必需功能基团.酶在2×10~(-4)M PMSF,活力仍不受影响,提示Ser残基与酶的催化功能无关.以Na_2S_4O_6保护游离巯基,用NBS修饰Trp残基,并辅以紫外光谱和萤光光谱分析.结果表明Trp残基为酶催化功能所必需.初步的CDC化学修饰结果亦表明,羧基似与酶催化活性有关.

Abstract:Actinidm was isolated and purified from fruits of wild Actinidia chinen-sis. The total number of SH groups of the enzyme molecule was estimated to be 7, and one of which has been shown to be the free SH group. By chemical modification with pCMB, it was found that the enzyme can be rapidly inactivated, suggesting that free SH group is an essential side chain in the active site of actinidin. Actindin preparation was selectively modified by NBS under the protection of Na2S4O6 and UV absorption spectrum and intrinsic fluorescence spectrum were conducted to demonstrate the situation of Trp residue in the enzyme molecule, the observed results suggest that Trp IS one of the essential groups of the enzyme.
