肝素－琼脂糖凝胶4B的制备及在肝生长因子提取纯化中的应用

以琼脂糖凝胶为载体，肝素为配基，乙睛活化后经环氧氯丙烷偶联，制得肝素——琼脂糖凝胶．经间接法测定，偶联率为２．４７５ｍｇ肝素／ｇ湿重琼脂糖凝胶，与Ｐｈａｒｍａｃｉａ公司的Ｈｅｐａｒｉｎ－ＳｅｐｈａｒｏｓｅＣＬ－６Ｂ比较，两种凝胶对肝细胞生长因子（ＨＧＦ）的亲和能力、层析行为及重复使用性能等方面非常相似，且自制凝胶的成本仅为进口的１／２０，解决了ＨＧＦ纯化过程中的关键步骤，使大量提取生产ＨＧＦ成为可能．

THE PREPARATION OF HEPARIN-AGAROSE GEL AND ITS APPLICATION IN THE PURIFICATION OF HGF

Using Sepharose 4B as ligand carrier and Heparin as the affinity ligand we prepaed the Heparin-Sepharose 4B with epichloronydrin as the cross-linker. The proportion of binding Heparin to Sepharose 4B is deterimoned by indirected method.The results demonstate that proportion of the binding Heparin to Sepharose 4B is 2.457 mg/g wet Sepharose 4B and the Heparin-Sepharose 4B is very similar to Heparin-Sepharose CL-6B made by Pharmacia LKB Company with the ability of binding HGF ,behavior of affinity chromatography and repeated-us-age.But the cost is only 1/20 that of Heparin-Sepharose CL-6B.So the procedures described here would be available for preparing large amount of HGF.
