摘    要:以硅胶干燥的白刺叶片为材料,分别在-84℃、-20℃冰箱保存和常温保存,对三种不同保存条件下的白刺叶片进行DNA的提取,结果表明,在-84℃和-20℃冰箱里保存1~2年的材料均可采用3×CTAB法有效去除多糖、酚及蛋白质,可获得高质量基因组DNA,而常温保存半年的材料无法获得DNA.通过进行ISSR—PCR实验,提取的DNA扩增均可得到稳定、清晰、多态性好的PCR扩增图谱,-84℃保存的材料所得到的DNA在ISSR—PCR扩增中条楷数目和清晰唐明昴优于-20℃保存的材料.

Abstract:The effects of total DNA extraction from Nitraria leaves dried with silica gel were studied under three different conditions for storing samples ( normal temperature and frozen at - 20 ℃and - 84℃ ) with improved 3 × CTAB method. It was suggested that the genomic DNA with high purity could be obtained after being frozen at - 20 ℃ or - 84 ℃ for one or two years. The polysaccharide, phenol and protein could be removed effectively. However the leaves at usual temperature could not provide the genomic DNA. Further results also showed that the leaf stored at - 84 ℃ was more suitable than the leaf stored at -20℃ for generating good ISSR-PCR banding patterns.
