| 摘 要： | 为了研究泛素连接酶Nedd4对肿瘤细胞增殖和凋亡的影响,利用小RNA干扰技术,通过慢病毒包装,侵染前列腺癌细胞DU145,达到下调Nedd4表达的目的.MTT检测表明,下调Nedd4表达可以抑制DU145细胞的增殖;DAPI染色发现,下调Nedd4表达的DU145细胞对星形孢菌素诱导的细胞凋亡更加敏感.这些结果都说明,下调Nedd4的表达不仅可以抑制DU145肿瘤细胞增殖,而且可以促进细胞凋亡.

| Abstract: | To study the effects of Nedd4 on tumor cell proliferation and apoptosis,Nedd4 was depleted in prostate cancer cell DU145 by infecting the lentiviruses expression of Nedd4-shRNAs. MTT assay showed that that depletion of Nedd4 de-creased DU145 cell proliferation. DAPI staining indicated that knockdown of Nedd4 resulted in greater sensitivity to stauro-sporine-induced apoptosis of DU145 cells. These results revealed that the down-regulation of Nedd4 expression not only reduced the cell proliferation but also induced the apoptosis of DU145 cells. |
