Relaxation effect of ethanol extracts from Croton crassifolius Geisel on mouse airway smooth muscle

To investigate the relaxation effect of ethanol extracts from Croton crassifolius Geisel (EECC) on airway smooth muscle of mouse. At tissue level， the effect of EECC on tracheal tension was detected by biological function system. At cell level， the effect of EECC on L-type voltage dependent calcium channels (VDLCCs) current was recorded by patch clamp system. In vivo， the effect of EECC on respiratory system resistance (Rrs) of mouse was detected by pulmonary function instrument. The results showed that at the tissue level， EECC dilated the trachea of mouse induced by high potassium or ACh in a dose-dependent manner， but had no effect on the normal trachea at rest. EECC blocked VDLCCs and other ion channels， thereby inhibited the calcium influx outside the cells and the calcium release in sarcoplasmic reticulum. At the cell level， EECC blocked VDLCCs current. In vivo， EECC significantly reduced the Rrs of mouse. The results suggest that the effective components of Croton crassifolius Geisel effectively relaxed the over contracted airway smooth muscle， and Croton crassifolius Geisel is expected to be used to alleviate or treat asthma.
