血管生长素衍生物His13Gly的基因工程研究

摘    要:在人工合成的血管生长素基因的基础上,利用化学合成的寡聚核苷酸片段,采用掺U法对ANG知性定位眯诱变,同位素杂交筛选到ANG基因的衍生物IS13Gl6突变子。

Abstract:On the base of chemically synthesizing angiogenic (ANG) gene, the highly expressing recombinant mutant-TG1/PBV220/ANG (rh') have been obtained by utilizing uracil-containing site-directed mutagenesis with mutagenic oligonucleotides and radiolabeled probe blot. Applied this mutant, the expressed angiogenin is about 36% of the total bacterial protein. After the purificatory steps of isolating inclusion body, utrasonic cleaning inclusion body, ion-exchange chromatography and molecular-sieve chromatography, the angiogenin purity was made up to 95%. Angiogenin was renatured by glutathione reoxidation. Applied embryo chroricallantoic membrane to examine the renature efficiency, the biological activity of competitively restrainging the wild type of angiogenin and the level of this activity.
