摘    要:以大肠杆菌为宿主表达了尖镰孢菌细胞色素P450 55A1(CYP450 55A1),并采用荧光光谱法研究了其与NO的相互作用.结果表明:在大肠杆菌(DE3)中成功表达了具有酶活性的CYP55A1.在280 nm波长激发下,NO的加入使CYP55A1的荧光逐渐降低,但随温度的变化差异较小;在413 nm波长激发下,NO的加入使CYP55A1的荧光逐渐升高,但加入NADH后其荧光又恢复到原来的值.

Abstract:Cytochrome P450 55A1(CYP55A1) from Fusarium oxysporum was expressed in E.coli, and the interaction between CYP55A1 and NO was studied by fluorescence spectrometry. The results indicated that CYP55A1 were expressed in E.coli (DE3) successfully. Under the excitation at 280 nm, the fluorescence intensity of CYP55A1 decreased gradually with the addition of NO but varied marginally with temperature. Under the excitation at 413 nm, the fluorescence intensity increased gradually with the addition of NO but would recover with the addition of NADH.
