| 摘 要： | 比较了4种毛白杨成熟叶片DNA的提取方法,结果表明,SDS法的提取效果优于CTAB法.其中,SDS-II法步骤较少,操作简便,不易造成实验误差,OD260/280值为1.87,提取的DNA片段长度在20kb左右,提取得率为42μg/g,为4种方法中最高.ISSR-PCR实验的效果表明,SDS-II法提取的DNA可以做为ISSR分子标记的模板DNA.

| Abstract: | The 4 DNA extracting methods for Populus tomentosa Carr.mature leaves were compared in this paper.The results show that the SDS methods are better than the CTAB methods.The author considers that the SDSII method is the best one with fewer steps,easy to handle and stable.The OD260/280 value of the SDSII method is 1.87 while the DNA yield rate is 42 μg/g.Agarose gel analysis shows the DNA molecular is 20 kb.The extraction is suitable acting as template DNA for ISSR-PCR amplification. |
