黑曲霉(UV—11)葡葡糖淀粉酶的分离纯化和酶的多型性性质的初步研究

摘    要:以黑曲霉UV—11的工业酶制剂为材料,分离纯化葡萄糖淀粉酶。蒸馏水抽提,硫酸铵盐析,凝胶过滤脱盐和DEAE一纤维素DE_(52)柱层析,得到4个部分(峰Ⅰ—Ⅳ),4个部份提纯0.92—1.84倍,酶活收率为26.6%。黑曲霉UV—11葡萄糖淀粉酶具有多型性,经聚丙烯酰胺凝胶水平板电泳共分出5条区带。粗酶和纯化的酶经电泳染色结果表明黑曲霉UV—11葡萄糖淀粉酶为一种糖蛋白。

Abstract:Glueoamylase was isolated and purified from commercial products of As pergillus niger mutant strains UV—11 has been shown to exist in multiple orms, seperable by polyacrylamide gel(7.5%)electrophoresis as nearly ocated bands. After ammonium sulfate fractionation at 60% saturated gel filtration (Sep hadexG—25)and DEAE—Cellulose DE_52 column chromatography, fou fractions(peak Ⅰ—Ⅳ)from the eluate were obtained.The four fractions have been purified 0.92—1.84 fold with a total recovery of 26.6%. Both protein and carbohydrate of crude enzyme and purified fractionsr were tested in PAGE, date showed that the Glucoamylase from A.niger mutant strains UV—11 was a glycoprotein.
