| 摘 要： | 以黑曲霉UV—11的工业酶制剂为材料,分离纯化葡萄糖淀粉酶。蒸馏水抽提,硫酸铵盐析,凝胶过滤脱盐和DEAE一纤维素DE_(52)柱层析,得到4个部分(峰Ⅰ—Ⅳ),4个部份提纯0.92—1.84倍,酶活收率为26.6％。黑曲霉UV—11葡萄糖淀粉酶具有多型性,经聚丙烯酰胺凝胶水平板电泳共分出5条区带。粗酶和纯化的酶经电泳染色结果表明黑曲霉UV—11葡萄糖淀粉酶为一种糖蛋白。

| Abstract: | Glueoamylase was isolated and purified from commercial products of As pergillus niger mutant strains UV—11 has been shown to exist in multiple orms, seperable by polyacrylamide gel(7.5％)electrophoresis as nearly ocated bands. After ammonium sulfate fractionation at 60％ saturated gel filtration (Sep hadexG—25)and DEAE—Cellulose DE_52 column chromatography, fou fractions(peak Ⅰ—Ⅳ)from the eluate were obtained.The four fractions have been purified 0.92—1.84 fold with a total recovery of 26.6％. Both protein and carbohydrate of crude enzyme and purified fractionsr were tested in PAGE, date showed that the Glucoamylase from A.niger mutant strains UV—11 was a glycoprotein. |
