枯草芽孢杆菌D-核糖摇瓶发酵条件的研究

摘    要:对枯草芽孢杆菌(Bacillus subtilis)S21进行了摇瓶条件下的D-核糖发酵条件的研究,主要考察了添加木糖、pH、溶氧水平以及采用补糖方式对发酵的影响,确定了D-核糖发酵的最佳工艺条件D-木糖50g/L初始pH7.0,初始葡萄糖浓度为150g/L,碳酸钙20g/L,并在发酵36h后,在装液量为35mL的500mL三角瓶中添加0.75g/mL的葡萄糖溶液2mL,S21菌可积累D-核糖达51.1g/L。

Abstract:Shake-flask culture conditions of D-ribose fermentation were studied with a Bacillus subtilis mutant S21.The paper investigated the factors affecting the yield of D-ribose,such as the amount of D-xylose in fermentation medium,pH、dissolved oxygen concentration and sugaring culture.It was suggested that the better fermentation conditon on laboratory scale was as following:D-xylose 50 g/L,initial pH7.0,initial glucose concentration 150 g/L,CaCO320 g/L.After it cultured 36 h,2 mL 0.75 g/mL D-glucose solution should be added to the 500 mL flask contained 35 mL fermentation medium.Under such optimized conditions,the yield of D-ribose was about 51.1 g/L.
