摘    要:利用CTAB法、SDS改进法和SDS快速法3种DNA提取代表性方法,对扩展青霉(Penicillium expansum)PF898基因组DNA的提取进行比较研究.CTAB法提取的DNA样品纯度最高,SDS裂解法制备的DNA得率最高;CTAB法和SDS改进法所得DNA都可直接应用于限制性酶切分析;3种方法所提取的DNA均能满足扩展青霉PF898脂肪酶基因和18S rRNA的PCR扩增.总体而言,SDS法简便、快速、得率高,是一种理想的扩展青霉基因组DNA提取方法.

Abstract:Genomic DNA of Penicillium expansum PF898 was prepared with three different methods(CTAB,modified SDS and rapid SDS method).The purity of the prepared DNA with CTAB was best.The modified SDS method produced the highest yield of DNA.The genomic DNAs prepared by both CTAB and the modified SDS were suitable for digestion with restriction enzyme.The samples of DNA by three method mentioned-above were suitable for molecular analysis on PCR for the gene encoding alkaline lipase from Penicillium expansum and 18S rRNA.To sum up,the modified method was ideal to prepare DNA from Penicillium expansum PF898 based on its simplicity,high purity and high yield.
