摘    要:本研究拟探索N-acetylgalactosaminyltransferase 14 (GALNT14)的表达水平对肾透明细胞癌(Kidney Renal Clear Cell Carcinoma,KIRC)的临床预后意义及其在疾病进展中的功能。从The Cancer Genome Atlas(TCGA)数据库获取肾透明细胞癌的临床信息和相应的mRNA表达数据,使用R语言进行差异表达分析和配对差异表达分析,检测GALNT14在肾透明细胞癌的表达;通过Logistic回归分析、Kaplan-Meier生存分析和COX回归分析探究GALNT14的表达量与肾透明细胞癌临床特征的相关性;从STRING数据库中获得GALNT14相互作用的蛋白质,通过Linkedomics和GeneMANIA数据库分析GALNT14的蛋白质-蛋白质相互关系;通过基因集富集分析(Gene Set Enrichment Analysis,GSEA)探索 GALNT14在肾透明细胞癌中介导的生物信号通路。结果表明,肾透明细胞癌组织中GALNT14的mRNA相对表达量显著高于癌旁组织;肾透明细胞癌GALNT14的高表达与肿瘤原发灶大小、Tumor Node Metastasis (TNM)分期、患者年龄、组织分级显著相关;肾透明细胞癌患者GALNT14的高表达与患者总生存率(Overall Survival,OS)正相关,与无病生存率(Disease Free Survival,DFS)负相关;GALNT14可作为肾透明细胞癌患者的独立预后因子;GALNT14的共调控基因与肾透明细胞癌关系密切;GALNT14的高表达与肿瘤细胞中氨基酸代谢、脂肪酸代谢和糖代谢等信号通路相关。因此,GALNT14在肾透明细胞癌中高表达且与患者预后相关,可作为该疾病的独立预后因子。

Abstract:The aim of this study was to investigate the prognostic significance of N-acetylgalactosaminyltransferase 14 (GALNT14) expression level in Kidney Renal Clear Cell Carcinoma (KIRC) and its function in disease progression. The clinical information and corresponding mRNA expression data of KIRC were obtained from The Cancer Genome Atlas (TCGA) database. The expression of GALNT14 in KIRC was detected by differential expression analysis and paired differential expression analysis using R language. Logistic regression analysis,Kaplan-Meier survival analysis and COX regression analysis were used to explore the correlation between GALNT14 expression and clinical characteristics of KIRC. Proteins interacting with GALNT14 were obtained from STRING database,and the protein-protein relationship of GALNT14 was analyzed by Linkedomics and GeneMANIA databases.The biological signaling pathway mediated by GALNT14 in KIRC was explored by Gene Set Enrichment Analysis (GSEA).The results showed that the relative expression of GALNT14 mRNA in KIRC tissues was significantly higher than that in adjacent tissues. The high expression of GALNT14 in KIRC is significantly correlated with tumor size,Tumor Node Metastasis (TNM) stage,patient age,and histological grade. The high expression of GALNT14 is positively correlated with Overall Survival (OS) and negatively correlated with Disease Free Survival (DFS) in patients with KIRC.GALNT14 can be used as an independent prognostic factor in patients with KIRC.The co-regulated gene of GALNT14 is closely related to KIRC. The high expression of GALNT14 is related to the signal pathways of amino acid metabolism,fatty acid metabolism and glucose metabolism in tumor cells. Therefore,GALNT14 was highly expressed in KIRC and is associated with the prognosis of patients,which can be used as an independent prognostic factor for this disease.
