油橄榄叶提取物联合美沙酮对海洛因成瘾大鼠的脑组织结构及单胺递质的影响

摘    要:目的:观察油橄榄叶提取物(olive leaf extract,OLE)联合美沙酮对海洛因依赖大鼠的脑组织结构及单胺递质的影响。方法:将28只健康大鼠随机分成正常组、模型组、OLE+美沙酮处理组和美沙酮处理组。按递增法采用皮下注射海洛因建立海洛因依赖大鼠模型,造模后药物处理30 d,在光学显微镜下观察大脑额叶皮层细胞结构的变化,利用荧光分光光度法检测脑中单胺类递质的含量。结果:与正常组相比,模型组海洛因依赖大鼠出现大脑额叶皮层结构不清、神经元发育不良、皮层厚度减小、脑单胺类递质含量明显增高;与模型组相比,OLE+美沙酮处理组和美沙酮处理组的大鼠大脑额叶皮层结构清晰、皮层厚度增加、脑单胺递质含量显著降低。结论:OLE联合美沙酮明显改善了海洛因依赖大鼠的戒断症状,保护了脑组织,其作用机制可能与脑内单胺递质的改变有关。

Abstract:Objective To investigate the effects of olive leaf extract( OLE) and methadone on brain histological structure and monoamine transmitters in the heroin dependence rats. Methods Twenty-eight rats were randomly divided into normal control group,model group,OLE and methadone treatment group and methadone treatment group. Each group of rats was subcutaneously given heroin with increasing dosage to establish heroin administration except the normal control group. After modeling,the rats were administrated with OLE and methadone for 30 d. The changes of the cell structure of cerebral frontal lobe cortex were observed by light microscope. The contents of monoamine transmitters were detected by fluorescence photometric method. Results Compared to the normal control group,The structure of cerebral frontal lobe was not clear,and the thickness of cerebral decreased. The contents of monoamine transmitters were increased significantly in the model group rats.Compared to model group,the structure of cerebral frontal lobe was clear,and the thickness of cerebral increased. The contents of monoamine transmitters were significantly reduced in treatment groups. ConclusionOLE and methadone in the treatment of heroin dependence rats can improve the withdrawal syndrome and protect the brain. These effects might be related with the changes of monoamine transmitters in the brain.
