摘    要: 蒜头果(Malania oleifera Chunet S.Lee)属于铁青树科(Olacaceae)蒜头果属.蒜头果蛋白(malanin)是通过粉碎、抽提、硫酸铵沉淀、疏水色谱层析等分离手段,从蒜头果种仁中分离、纯化出的一种新的植物蛋白.Malanin是一种糖蛋白,分子质量为61875u,等电点为5.5,由A,B2条肽链通过二硫键连接而成.该蛋白的A链和B链N-端序列分别为DYPKLTFTTS和DETXTDEEFN(X一般情况下为C),用NCBI-BLASTP程序对nr数据库和其它蛋白质数据库进行检索,未发现有相同的序列存在.经基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)及分析各自的肽质量指纹图谱(PMF),结果没有匹配数据库中的任何记录,说明malanin为新蛋白质.

Abstract:Malania oleifera Chun et S.Lee belongs to Olacaceae.Anovel protein,designated as malanin,was isolated and purified from the seeds of Malania oleifera by homogenization,extraction with phosphate solution,followed by ammonium sulfate precipitation and hydrophobic interaction chromatography.Malanin is a glycoprotein with a molecular weight of 61875 Du and an isoelectric point of 5.5,and it was linked with two bands of A-chain and B-chain by one or more disulfide bonds.The N-terminal amino acid sequences of malanin are DYPKLTFTTSin chain-A,and DETXTDEEFN(Xwas commonly C) in chain-B.The N-terminal sequences of A-chain and B-chain had been blasted by NCBI-BLASTPand other databases with the result of no significant similarity.MALDI-TOF-MS methods were applied to measure the peptide mass fingerprinting(PMF) of malanin-Aand malanin-B,but no significant protein similarity was found by the protein database of Peptldent,suggesting that malanin is a novel protein.
