用紫斑牡丹不同外植体诱导愈伤组织的研究

摘    要:先后分别用紫斑牡丹(Paeoniarockii)的土芽、叶柄、叶片、幼芽、心皮、雄及经一段时间组织培养的试管苗等作外值体,进行愈伤组织的诱导。结果表明,配方MS+BA1.5mg/L+NAA2mg/L最为理想,不同外植体中,试管苗的幼芽和土芽诱导率最高,叶柄和叶片次之,其诱导率分别为100%,60%,30.8%;而生殖器官雄和心皮未能诱导成功,在上述基础上讨论了牡丹愈伤组织的诱导形成与组织分化水平和类群进行水平的关系。

Abstract:The callus have been induced on MS medium added into different density of BA and MAA with petiole,balade,tender bud,stamen and pistil as explant respectively.The results indicate that MS BA?1 5?mg/L NAA?2?mg/L is most suitable for forming callus.The tender bud of test tube plant is the best among different explant.Petiole and blade are no less than it,the induction rates of callus are 100% for slander bud,60% for petiole,30 8% for blade.But reproductive organs were not successful for callus induction.The relationship among evolutionary level of taxo and induction of callus as well as dedifferentiation level of tissue have been discussed on the basis of induction of callus.
