草履虫接合生殖标本的高密度富集与快速制片技术研究

摘    要:针对传统方法中,收集的草履虫接合生殖材料样本少、固定时材料易收缩、固定后的材料不易保存等限制,突破性地采用初固定、再固定两步固定方式,及使用不同目数的特制滤器进行富集和纯化,一次性地获得大量的高密度草履虫接合生殖样本,可高达80%~90%.固定、清洗、染色、脱水、透明等制片过程均在滤器中进行,无须离心机离心等传统处理,透明后的材料直接浸润、保存在液体封固胶内,可长期保存,随时取材,随时制片,为教学、科研、制片带来前所未有的极大方便。

Abstract:According to traditional methods, it was difficult to enrich conjugation sample of Paramecium, and the material was fragile to shrink when being fixed. What's more, it was impossible to conserve the sample well after it was fixed. On account of these limits, we renovated the methods of enriching the sample. A substantial amount of Conjugation sample of Paramecium was obtained in this paper by means of enrichment or purification through a specific stepwise filter. A series of steps including fixation, dyeing, washing, dehydration and clarity could be finished in the filter instead of in a centrifuge. The material after clarity could be soaked and conserved in liquid gel for a long time. You may take the material whenever you like, which facilitate to both teaching and research.
