皮燕麦胚性愈伤组织的形成及植株再生

摘    要:本实验以皮燕麦2079 成熟种子胚为外植体,其愈伤组织的诱导率最高,叶鞘愈伤组织诱导率为5 % ,叶片几乎不发生脱分化。在基本培养基比较中以改良N6 培养基诱导效果为佳,愈伤组织出愈率在1-3mg/L,2 ,4—D浓度范围逐渐提高,继续增大时则出愈率降低。胚性愈伤组织形成率受脱分化培养基中2,4 —D浓度的影响。扫描电镜观察结果显示非胚性与胚性愈伤组织表面结构差异明显;组织切片表明形成的再生植株有胚胎发生与器官发生两种途径,形态上有区别。

Abstract:The efficiency of callus induction was much higher,if mature embryos of oats were used as explants.No dedifferentiation was observed by using leaf blade as explants.The best medium was modified N 6.The efficiency of callus raise when 2,4-D concentration increased from 1 to 3 mg/L.Overhigh concentration of 2,4_D was worse.The concentration of 2,4-D in dedifferentiation medium had relation to embryogenie callus formation.It was different bettew the surface structure of the embryogenic and non-embryogenic calluses by means of observation by scanning electron microscope.The callus sections showed That the Plantlet regeneration was differentiated ways.
