| 摘 要： | 利用SDS高盐法对冰川微生物总DNA进行了提取，实验结果表明这种方法对不同区域的冰川都可以提取到长度大于15kb的DNA片段，所得DNA应用细菌16S rRNA基因的引物进行PCR扩增，均能获得目的片段。因此，SDS高盐法是一种高效、简单的从小量冰川样品中直接提取DNA的理想方法。

| Abstract: | In this paper, glacier microbial DNA was extracted by the method of SDS lysis, The result showed that this method could extract DNA with more than 15kb in size from different areas. They were used as templates for PCR amplification with success, using primers of bacterial 16S rRNA gene. Therefore, method of SDS lysis is more suitable for DNA extraction directly from a small amount of glacier sample. |
