转Bar基因抗除草剂稻谷对小鼠致敏性的研究

摘    要:为研究转Bar基因抗除草剂稻谷是否具有致敏性,采用SDAP、Farrp和NCBI三大数据库对磷丝菌素乙酰转移酶(PAT)进行致敏原序列对比,并用蛋白质结构模拟SWISS-MODEL预测PAT的三维构象并对其进行分析.将60只昆明小鼠分为2组,分别喂养含相同剂量Bar基因水稻和常规水稻90 d,持续3代,每组抽样20只用ELISA分别检测其肠道粘液免疫球蛋白A(sIgA)、血清二胺氧化酶(DAO)和血清免疫球蛋白E(IgE).蛋白质生物信息学比对结果表明,PAT酶与数据库中已知致敏原无任何同源性,三维构象亦表明其与其他N-乙酰转移酶大家族(NATSF)成员相似,无致敏性的可能;ELISA的3项检测指标均无显著性差异(P>0.05).结论:在本试验条件下,转Bar基因的稻谷对小鼠无明显致敏性.

Abstract:To study the potential allergenicity of the Bar-GM rice,database SDAP,Farrp and NCBI were used in protein sequence alignment of PAT,and the three-dimensional conformation of PAT was formed and analysed in SWISS-MODEL.60 KM mice were divided randomly into two groups,rationed respectively by the feed contains Bar-GM rice and the same strain normal rice in the same dose.After 90 days,20 mice of each group were sampled at random to detect the IgE and DAO content in their blood serums and sIgA in their small intestines mucus with ELISA,lasting for three generations.There was no positive match through the protein-bioinformatics comparison,PAT had no homology with any of the known allergens.The three-dimensional conformation inspection indicated that PAT had a similar structure with other members in NAT-SF.There was little possibility to cause anaphylactic reaction.The three test indexes of ELISA all showed no significant difference between the two groups(P>0.05).From all of the above tests,a conclusion can be drawn that Bar-GM rice has no obvious allegenicity.
