磁性适配体纳米荧光生物传感器信号增强检测腺苷(英文)

摘    要:研制了一种简单的磁性适配体纳米荧光生物传感器用于信号增强检测腺苷.同另一条荧光标记的DNA (FAM-sDNA)部分互补的腺苷适配体首先被固定在金包磁纳米颗粒上,当加入腺苷时,由于腺苷与其适配体的亲和力大于双链互补力,FAM-sDNA从传感器上解离,磁性分离之后,再将增敏剂乙醇加入溶液中,当腺苷浓度在8.7×10-5~3.4×10-3 mol·L-1范围内,荧光信号与腺苷浓度呈较好的线性关系,检测限达到7.0×10-6mol·L-1.该方法简单、快速、灵敏、且具有良好的选择性,在小分子检测和疾病诊断中具有应用潜力.

Abstract:A simple fluorescence signal enhancement method for highly sensitive detection of adenosine based on magnetic aptamer nano-flares biosensor(MANFB) was developed.The biosensor was fabricated by immobilizing a flare strand,a fluorescein labeled oligonucleotides(FAM-sDNA) partly com-plemented with adenosine-aptamer on Aucoated magnetic nanoparticles(AuMNPs).The flare strand assignal source and ethanol as enhancement agents were used for high sensitive detection of adenosine.In the presence of adenosine,the fluorescein labeled oligonucleotides(FAM-sDNA) was released from theMANFB because of the combination between the target and its aptamer.After magnetism separation,ethanol is further added to the solution to enhance the fluorescent signal.The fluorescence intensity linearresponse to adenosine concentration in the range of 8.7×10-5~3.4×10-3 mol·L-1 and the lowest detection limit down to 7.0×10-6 mol·L-1(S/N =3) was obtained with the proposed method.The method proposed in this work can be used for simple,rapid,sensitive and highly specific adenosine detection,and it shows promising potential in small molecules detection and disease diagnosis.
