正交设计在黄连中小檗碱提取实验教学中的应用

摘    要:通过正交表的结构和选用、因子和水平的合理选择等教学活动,把正交实验法应用于黄连中小檗碱提取的学生实验,经过教师引导,学生参与讨论,确定了稀硫酸浓度,稀硫酸的用量、石灰乳调碱度的pH和趁热抽港后用盐酸调pH的温度为提取的因子,每个因子三个水平,选用了L9(34)作为正交实验表,学生实验后,通过直观分析和方差分析,得出此次实验最佳方案为:稀硫酸以16倍量,浓度为0.2%,浸泡24小时。石灰乳调碱度的pH为9,盐酸调pH的温度为60℃。

Abstract:Based on the construction and choice of orthogonal array,on the selection of factors and levels, orthogonal experiment for the students is applied to the extraction of berberine in the Rhizoma Coptidis. The concentration and the capacity of sulfuric acid, the regulation of pH by Ca(OH)2 and the decision of the temperature when the acidity is regulated by hydrochloric acid after being filtered are choosed as the factors of the extraction. Each factor has three levels. Orthogonal layout of Lg(34)is choosed. After the experiment, the best program is reached by a direct and variance analysis. 16 times and 0.2% sulfuric acid is choosed. The powder of Rhizoma Coptidis is soaked for 24 hours.pH is regulated to nine by Ca(OH)z. The temperature when the acidity is regulated by hydrochloric acid is 60℃.
