泰山四叶参多糖的提取、含量测定及抗氧化活性研究

摘    要:本实验研究泰山四叶参多糖的提取、分离、纯化方法,并测其含量,探讨其抗氧化能力.方法是建立热水提取、乙醇沉淀、Sevage法除蛋白及透析、纯化泰山四叶参多糖的方法,采用苯酚-浓硫酸法测定其含量,并比较了不同浓度的四叶参多糖(200、400、600、800、100μg/ml)和抗坏血酸(Vc50、100、200、400、600、800、100μg/ml)的还原能力以及不同浓度的泰山四叶参多糖和抗坏血酸(均为40、80、160、320、600、800μg/ml)清除二苯代苦味酰基(DPPH)自由基的能力.结果显示,本实验提取得到泰山四叶参多糖含量为11.09%,泰山四叶参多糖的还原能力及清除DPPH自由基的能力随其浓度的增加而增大,在800μg/ml时,DPPH自由基清除率达到90%以上,接近抗坏血酸的清除率.因此,泰山四叶参多糖具有较好的抗氧化能力.

Abstract:Objective: To research the method of the extraction,determination and antioxidant capacity of Polysaccharides from Radix Codonopsis Lanceolatae(RCLP) from Mount Taishan.Methods: Polysaccharides was extracted with hot water,subsided with 95% ethanol,separated with protein with Sevage assay and then dialyzed and purified.The content of RCLP was assayed with phenol vitriolic colorimetry.We determinate and compare the deoxidization activities of RCLP(200,400,600,800,1000μg/ml) and vitamin C(50,100,200,400,600,800,1000μg/ml) and clearing DPPH free radical activities of RCLP and vitamin C(40,80,160,320,600,800μg/ml).Result: With augmentation of concentration,the deoxidization and scavenging DPPH free radical capacity of RCLP increased continuously.RCLP(800μg/ml) could clear 90% DPPH free radical.That was close to Vc.Conclusion: RCLP from Mount Taishan has relatively strong antioxidant activity.
