摘    要:以自制的洋葱伯克霍尔德菌固定化脂肪酶为催化剂,在微水相、无溶剂体系中研究了大豆油和甲醇合成生物柴油的工艺。在系统考察了酶用量、醇油比、含水量、反应温度、反应时间、甲醇流加方式等因素对甲酯得率影响的基础上,利用响应面试验设计优化了各主效因子,建立甲酯得率的二次回归方程,获得了最优的工艺条件:加酶量2.4%、加水量7.1%、反应温度40.4℃、反应时间10.7h、 醇油比4.5。在此条件下,实验测得甲酯得率为97.2%,与响应面模型预测值96.9%非常吻合,说明该优化方法有效、可靠。

Optimization of the production of biodiesel catalyzed by immobilized lipase from Burkholderia cepacia using a response surface method

Abstract:Lipase from Burkholderia cepacia has been immobilized and used for the production of biodiesel from soybean oil and methanol in a microaqueous and solvent-free system. The effects of the enzyme loading, water content, reaction temperature, reaction time, methanol/oil molar ratio and number of times of adding methanol on the yield of methyl ester were investigated in detail, and the factors having a major impact were optimized by a response surface method. A quadratic regression equation for the yield of methyl ester was then established. The results showed that the optimum conditions were an enzyme loading of 2.4%, water content of 7.1%, reaction temperature of 40.4℃, reaction time of 10.7 h, and methanol/oil molar ratio of 4.5. Under these conditions, the measured yield of methyl ester was 97.2%, which was very close to the predicted value of 96.9%. This indicates that the optimization method is effective and reliable.
