| 摘 要： | 应用非离子型去垢剂Brij58增溶牛精细胞获得的膜蛋白,经VBL-Sepharose4B亲和柱层析分离,可获得在PAGE、SDS-PAGE中均呈现单一蛋白带的VBL受体.其亚基分子最为46500d.牛精细胞经Brij58增溶的膜蛋白经用~(125)I标记,再经VBL-Sepharose4B亲和柱层析,也能获得~(125)标记的凝集素受体.经测定,用亲和层析法获得的凝集素受体,保持了原有的生物学活性.

| Abstract: | Bovine spermatozoa were treated with non-ionic detergent and the excessive detergent in soluoilizing extract can be adsorbed and eliminated by ion-exchange resin, then, the VBL receptors in plasma membrane composition sclubilized from bovine spermatozoa were separated by affinity chromategraphy on a column of VEL-Sepharose 4B. The purified VBL receptor yield one protein band on SDS-PAGE, corresponding to a subunit molecular weight of 46,5000 dalton.The composition solubilized from bovine spermatezca by Frij-58 was labeled by 125I, then, separated by affinity chromatography on a column of VBL-Sepharose 4B and obtained a 125I-receptor of VBL.The purified receptor can be determined by the agglutinating activity's inhibition or declining after receptors bind with VBL, or by studing 125I-VBL's binding with the receptors. The binding of receptor with lectih can be inhibited by D-Man All these results prove that the purified receptor have natural biological activity. |
