PSⅡ反应中心复合物制备条件的选择

摘    要:采用Triton X—100处理、超速离心和离子交换层析等方法,由菠菜PSⅡ颗粒制备得PSⅡ反应中心DI/D2/Cyt b559复合物。整个过程分为PSⅡ颗粒的解体和解体后反应中心的分离两步骤。避光、低温、PH值、解体时间和Triton X—100与PsⅡ颗粒的相对量是影响PSⅡ颗粒解体的因素。Triton X—100浓度和NaCl浓度则影响解体后反应中心复合物的分离。并比较了去垢剂类型、植物种类和叶片生长期对PSⅡ颗粒解体的影响.

Abstract:A photosystem II reaction center D1 -D2 -Cyt. b559 complex was isolated from spinach PS n particles by extracting with Triton X-100, followed by ultracentrifugation and ion - exchange chromatography with DEAE-Toyopearl 650S. The various conditions of preparasion were compared. The results show that severial factors affect the preparation of PS II RC. Among these,the effects of the time of Triton X-100 extraction and the pH valus of sample buffer are the most notable, and the effect of the ionic strength of buffer is more notable. The effect of the extract and disassociation of several kinds of detergents for PS II particles was discussed.
