维医证候异常黑胆质动物模型微循环状态的改变

摘    要:为探讨异常黑胆质证候动物模型微循环状态的改变,选择健康ICR雄性小鼠30只,分为模型组和正常对照组,每组15只。正常对照组和模型组造模后的实验动物眼球放血处死,并迅速取出舌和前右侧趾,一部分采用石蜡切片苏木精-伊红(HE)染色,光学显微镜下进行组织学观察;另一部分采用透射电镜观察细胞超微结构改变。采用对比研究的方法观察和比较两组实验动物微循环状态的改变。结果显示,与正常对照组相比,模型组小鼠舌和甲床组织病理学有明显地改变和细胞器损伤。光镜下观察,模型组蕈壮乳头和上皮下结缔组织内微血管扩张、淤血,血管周围偶见少量炎性细胞;甲床真皮浅层血管与骨髓内血管扩张、充血。电镜下观察,模型组毛细血管管腔狭窄,呈不规则状,管壁粗糙,内皮细胞异染色质增多;甲床毛细血管内皮细胞异染色质明显增多。由此得出结论:异常黑胆质的产生过程中机体有微循环状态的改变。

Abstract:In order to study the changes of microcirculation in animal model for abnormal savda syndrome,30 healthy ICR(Institute of Cancer Research) male mice were chosen and divided into the model group(15) and the control group(15),respectively.Experimental mice were executed by eyeball blood letting and the tongue and ante-right side toe of the mice were taken out immediately.One part of the tissue was treated by HE staining and observed under the optical microscope in order to analyze the histology.The other part was observed by TEM(Transmission Electron Microscope) to analyze cell ultra -structural changes.A comparative method was used to observe and compare the microcirculation changes of the two groups.The results indicate that as compared to the control group,the model group sees obvious histopathology changes and organelle damages in tongue and nail bed.Light microscope observations reveal vasodilatation,congestion inside the mushroom nipple and the subcutaneous connective tissue in the model group.Some inflammatory cells were observed on the surrounding blood vessels wall;and also vasodilatation and hyperemia in nail bed dermis superficial layer blood vessel and inside the marrow.Under electron microscope observation,the blood capillary lumens were found to be narrow and irregular;vessel wall to be rough in the model group,endothelial cell saw multiplications in heterochromatin;nail bed capillary endothelium cell saw obvious increase in heterochromatin.It may be concluded that there is microcirculation disturbance during the occurrence of abnormal savda.
