Survivin基因在胃癌组织中的表达及其与凋亡的相关性研究

目的研究胃癌组织中Survivin基因的表达及其与凋亡的关系。方法应用免疫组织化学SP法检测30例正常胃粘膜及120例胃癌组织中Survivin基因的表达；采用脱氧核糖核酸末端转移酶介导的缺口末端标记（TUNEL）技术，检测其凋亡指数。结果30例正常胃粘膜组织Survivin基因均不表达，胃癌组织中Survivin阳性率为62．5％（75／120），二者有显著性差异（P〈0．01）；Survivin基因表达与组织分化程度和TNM分期密切相关（P〈0．05）。胃癌组织凋亡指数（AI）和TNM分期、组织分化程度密切相关（P〈0．05）。Survivin基因表达水平与凋亡指数呈显著负相关（P〈0．01）。结论Survivin基因在胃癌的发生发展过程中可能起重要作用。Survivin基因的表达可抑制胃癌细胞凋亡、促进胃癌细胞增殖。

Expression of Survivin with Apoptosis in gene and its relationship Gastric Carcinoma

Objective To study the expression of Survivin gene in gastric carcinoma and the relationship between Survivin expression and apoptosis in the tumor. Methods The expression of Survivin was examined with immunohistochernical S - P method in 30 specimens of normal gastric mucosa and 120 specimen of gastric carcinoma. The apoptosis index（AI） was examined with terminal deoxymueleotidyl transferase mediated dUTP nick end labeling（TUNEL） technique. Results All nonnal mueosas were negative for Survivin. In the 120 gastric carcinomas. 62.5 % （75/120） were postive for Survivin, The expression of Survivin was well associated with histological differentiation and TNM stage（ P 〈 0.05）. The apoptosis index（AI） of gastric carcinoma cell was negatively associated with TNM stage and histological differentiation（ P 〈 0.01 ）. The expression of Survivin was negatively correlated with the AI of gastric carcinoma cell（ P 〈 0.01 ）. Conclusion Survivin was overexpressed in gastric cancer. Survivin gene may play an important role in the course of tumorigenesis and progression in gastric carcinoma. Survivin gene may inhibit apoptosis of gastric cancer cells, and increase cell proliferation. It may be valuable to evaluate the malignant degree of gastric carcinoma. The inhibition of apoptosis in gastric carcinoma could related with the up - expression of Survivin.
