c-fos在吸烟刺激骨吸收中的作用

摘    要:目的探讨吸烟对人骨组织c—fos基因表达的影响。方法选取吸烟和不吸烟健康成年志愿者各10名,分为吸烟组和非吸烟组,吸烟组患者为每天吸烟20支以上,烟龄不少于10年;非吸烟组患者为不吸烟或每天吸烟不超过1支。各取约1克髂骨松质骨,提取其总RNA和总蛋白,采用RT—PCR测定C~fos mRNA的表达:采用Western blotting测定fos蛋白表达;采用免疫沉淀法测定fos蛋白磷酸化程度。结果吸烟组RT-PCR条带光密度值的c—fos/β-Actin比值高于非吸烟组的比值,差异有统计学意义(P〈0.05)。Western blotting检测,吸烟组c~fos的条带光密度值高于非吸烟组,差异有统计学意义(P〈0.01)。吸烟组c—fos免疫沉淀的条带光密度值高于非吸烟组,差异有统计学意义(P〈0.01)。结论吸烟可以导致人骨组织c—fos基因与c—fos表达增加,其在骨质疏松发生中的作用有待进一步研究.

Effect of smoking on the expression of both c - fos mRNA and c- fos in human bone tissue

Abstract:Objective To explore the influence of smoking on the expression of both c - fos mRNA and fos in human bone tissue. Methods Twenty healthy adult volunteers had been divided into two groups. Out of the 20 volunteers, ten had used to smoking above 20 cigarettes at least 10 years daily, and the ten cases had made up of smoking group. The others had used to no smoking or smoking no more than 1 cigarette daily, and them has made up of non - smoking group. Each of them had been taken out about 1 g cancellous bone in ilium. RTPCR and Western blot had been performed to detect the expression of c - fos mRNA and protein respectively. The c - los protein phosphorylation degree had been detected by immuno- precipitation. Results The e - fos/betaAetin ratio of band optical density values in smoking group had been higher than in non- smoking group basing on the RT- PCR amplification, and the differences had statistical significance( P 〈 0.05). The c - fos band optical density values in smoking group had been higher than in non - smoking group basing on the western blotting analysis, and the differences had statistical significance( P 〈 0.05). The c - fos band optical density values in smoking group had been higher than in non- smoking group basing on the irnmuno-precipitation analysis, and the differences had statistical significance( P 〈 0.05). Both the level of c - fos mRNA and c - fos had been higher markedly in smoking group than in non- smoking group, the difference had statistical significance( P 〈 0.05). Conclusion Smoking up - regulates the expression of both the c - los mRNA and the c - los in human bone tissue, whether it is involved in the process of osteoporosis or not awaits further study.
