摘    要:目的:探讨合成含CpG基序的寡核苷酸(CpG ODN)对重组乙型肝炎表面抗原(rHBsAg )增强小鼠特异性抗体产生及淋巴细胞增殖反应的效应.方法:采用BaIb/c小鼠作为免疫实验动物,经后腿胫骨前肌免疫2次,ELISA法检测血清乙型肝炎表面抗体(抗-HBs)效价;3H-TdR掺入法测定淋巴细胞增殖反应.结果:加CpG ODN组的抗-HBs效价较rHBsAg组明显增高,且维持时间长;淋巴细胞增殖反应显著增强.结论:CpG ODN对小鼠抗-HBs产生具有明显的增强作用,且可以有效刺激小鼠淋巴细胞的增殖反应.

Abstract:To investigate effects of synthetic oligodeoxynucleotide containing CpG motif (CpG ODN) on specific antibody production and lymphocyte proliferation to recombinant hepatitis B surface antigen (rHBsAg). Methods: Balb/c mice received into back leg tibialis anterior (TA) muscle for two times;Detection of anti-HBs titer in serum by ELISA;The lymphocyte proliferations are tested by 3H-TdR incorporation. Results: The anti-HBs titer of plus CpG ODN group is higher than that of rHBsAg group obviously, and the antibody persistent time is also longer. Nevertheless, the proliferation of splenocytes of the group plus CpG ODN is much stronger. Conclusion: CpG ODN has enhanced efficacy for anti-HBs production and stimulated the splenocytes proliferation in mice.
