淡水无核珍珠壳角蛋白的酶解及其色氨酸分析

摘    要:本文试验用胰蛋白酶,链丝菌蛋白酶,木瓜蛋白酶对从三角帆蚌珍珠及其蚌壳珍珠层中提取的壳角蛋白进行酶法水解,采取荧光法对壳角蛋白酶解液进行色氨酸(Trp)含量及酶解程度的分析检测。结果表明:其酶解液和空白酶液在280nm激光波长下都具有单一的350nmTrp所特有的荧光发射峰,但酶解液的荧光强度明显大于空白酶液荧光强度,从而说明珍珠及其贝壳中Trp的存在。通过定量分析,壳角蛋白中Trp含量大约在2.8

Abstract:The experiment in this article takes enzymolysis to Conchiolin which is abstracted from Anoclonta woodianalea and its pearl layer with Trypsisn pronase and papain etc. ,Then makes Trytophan analysis and hydrolytic test to Enzymic hydrolysate of Conchiolin by fluorometry method. These results shows: Enzymic hydrolysate and Pure Enzyme liquid have a unitary fluorometry emission peak of 350nm which is what Trytophan only has at 280nm excited wavelength,but Fluorescence of Enzymolysis liquid is more clear than that of Pure Enzyme liquid. So, from it, we know that there exists Trytophan in pearl and its shell layer. By quantitative analysis, the Trp content in Conchiolin is about 2. 8 - 3. 5%. The experiment also shows :Trypsin,Prnase and Papain can enzymoly Conchiolin,and the effect of Trypsin higher than that of the others.
