摘    要:为了进一步认识拟穴青蟹一氧化氮合成酶(S.paramamosain nitric oxide synthetase,SpNOS)蛋白的结构和功能,深入研究该蛋白的生物学特性,本研究采用生物信息学方法,对SpNOS蛋白质的理化性质、二级结构及三级结构等进行生物信息学分析,并在获得三级结构的基础上进行同源建模.分析结果表明,拟穴青蟹NOS蛋白与眼斑龙虾NOS蛋白组成较为接近.二级结构以α螺旋和随机卷曲为主.NOS全长1214个氨基酸,其空间结构分别以2G60、3HR4、1QGY的A链为模板,分三段进行建模,得到合理的空间结构.实验数据为深入研究一氧化氮合成酶的蛋白结构和免疫功能的关系奠定了基础.

Structure Analysis and Homology Modeling of NOS Protein from Mud Crab Scylla Paramamosain
Abstract:A series of bio-informatics analyses were carried out to investigate the potential characteristics and structure of the NOS(Nitirc Oxide Synthetase) protein from a 1214-aa-long sequence obtained from mud crab S.paramamosain.The result of general property of S.paramamosain NOS showed that it had the highest similarity with NOS from Palinurus argus in terms of amino acid composition.The secondary structure was predicted and showed that α-helices and random coils were main elements in the molecule NOS.As amino acid chain of the NOS was too long,a three-dimensional structure was established based on the chain A of 2G60,3HR4 and 1QGY.The result of homolog modeling is proved to be reliable by Ramachandran plots.Further investigation is proposed to show the relationship between the function of NOS in the immune system and the structure of NOS in S.paramamosain.
