米曲霉与黑曲霉原生质体种间电融合杂交育种

摘    要:研究了培养基、菌龄、缓冲系统、渗稳剂、酶系统等因素对米曲霉和黑曲霉原生质体制备和再生的影响,探讨了电融合的最佳条件。通过种间原生质体电融合,得到了稳定的重组子。测定了融合子的生长速度、蛋白酶活性和酯酶同工酶酶谱,与亲本进行比较,得到两株生长较快、蛋白质酶活性较高的融合菌株,拮抗实验表明,他们是不同于亲本菌的新菌种。

Abstract:his paper investigates the effects of factors such as cultivation media,mycelial ages, buffer systems,osmatic stabilizers and enzyme systems on the formationand regeneration of protoplasts of A. niger and A. oryzae and the conditions of protoplastelectrofusion and obtains stable fusants by means of interspecific electrofusion. Thegrowing speed, protease activity and esterase isoenzymes of the fusants were measured andcompared with their parents. Two fusants with relatively rapid growing speed and highactivity of protease were obtained. The colony antagonism test shows that they are newstrains different from their auxotrophic parents.
