MALDI-TOF MS 测定多肽和蛋白质混合物的可行性研究

摘    要:考察了基体辅助激光解吸电离飞行时间质谱 (matrixassistedlaserdesorption ionizationtime_of_flightmassspectrometry,MALDI_TOFMS)测定多肽和蛋白质混合物的可行性。实验结果表明 :MALDI_TOFMS能快速地同时提供多肽和蛋白质混合物中各组分的准确相对分子质量。对于相对分子质量较小的多肽混合物 ,采用反射模式 ,多肽组分分子离子峰M H 的各同位素峰清楚地区别开来 ,达到了基线分离水平。对于 5种相对分子质量在10 4 6 5 4~ 6 6 4 31 0 0范围内的多肽和蛋白质混合物 ,采用线性检测模式 ,MALDI_TOFMS同样能够检测到各组分的分子离子。

Abstract:The possibility of analysis of peptides and proteins mixtures by matrix_assisted laser desorption/ionization time_of_flight mass spectrometry (MALDI_TOF MS) was studied. The results showed that this method can detect molecular ions of the peptides and proteins in the mixed samples. Under reflex mode,isotopeaks of molecular ion peaks for all \{peptides \} were clearly discriminated on the base line level. Under liner mode,molecular ions of mixed peptides and \{proteins\} also were detected and distinguished clearly.
