刺桐胰蛋白酶抑制剂亲和层析填料的制备及应用

摘    要:研究用交联琼脂糖为载体,三聚氯氰为活化剂,刺桐胰蛋白酶抑制剂为配基,制备亲和填料的方法.通过紫外和红外光谱分析,对制备的填料进行表征.用制备的亲和填料吸附重组人组织型纤溶酶原激活剂突变体(reteplase,r-PA),研究吸附与解吸附条件.结果表明,用该方法制备的亲和填料,制备工艺简单,吸附性能良好,对r-PA的吸附量为8.76 mg·g-1.解吸附完全,不影响活性,可以用来分离纯化r-PA.

Abstract:Taking sepharose 4B as carrier, cyanuric chloride as activation reagent, and Erythrina trypsin inhibitor as ligand, the method of preparation of affinity chromatography resin is described. The UV and IR spectral properties of the resin are studied. The resin is used for adsorbing r-PA. The factors influencing the adsorption and desorption are investigated. The result indicates that the procedure of preparation is simple, and the adsorption capacity is high. The highest capacity of the resin to r-PA is 8.76 mg·g-1. The adsorbed protein on the resin can be eluted adequately and the activity of the protein has no loss. The resin can be used for the separation and purification of r-PA.
