蛋白质晶体重原子衍生物衍射强度的统计公式

为解决测定胰岛素晶体结构中所出现的问题,作者曾导出重原子衍生物X射线衍射强度的统计公式,用来计算衍射强度的相对比例系数K_(pH)/K_p。此项方法于1968年秋提出,深受北京大学周公度同志和唐有祺教授的支持,并于1969年初应用于胰岛素晶体结构测定的研究工作,且得到梁栋材同志及参加工作的同志的全力支持。 继胰岛素晶体结构研究工作之后,我国又接着开展天花粉晶体结构的研究工作。因此,发表这一研究简报,有助于兄弟单位相互借鉴,少走弯路。

STATISTICAL FORMULAS FOR X-RAY DIFFRACTION INTENSITIES FOR HEAVY-ATOM DERIVATIVES OF PROTEIN CRYSTALS

Using statistical methods, two equations for isomorphous heavy-atom derivatives of protein cyrstals can be obtained: in the case .In the derivation of formulas (1) and (2) we have avoided using diffraction data of anomalous dispersion owing to their relatively large errors in measurements. The quality of diffraction data and isomorphous degree may be analysed primarily by these formulas. The obtained values of the above scale can be used to improve the convergence in the Least Square method for the refinement of heavy-atom parameters of protein crystals. Formulas (1) and (2) have been tested and used in determination of the insulin cyrstal structure.
