烟草多酚提取工艺优化及其主要成分定性分析

摘    要:采用微波辅助技术提取烟草多酚,利用质谱分析及二级管阵列光谱扫描(HPLC-PDA-MS)对烟草多酚主要组分定性分析。研究了烟草多酚提取条件和烟草多酚主要组成成分。经响应面优化确定烟草多酚的微波提取条件为:甲醇浓度53%,料液比1:12,微波功率510W,提取时间90s,在此条件下总多酚的提取得率为27.21mg/g(干重)。HPLC-PDA-MS定性烟草多酚主要组分为绿原酸和芸香苷。

Abstract:The tobacco polyphenols were extracted by microwave treatment, the main component in tobacco polyphenols was identified by HPLC-PDA-MS. Effects of microwave power, concentration of methanol ,extraction time and ratio of liquid to material on extraction yield were investigated by using microwave technology, and then the experiment of response surface analysis (RSA) was designed. The optimum extraction conditions for tobacco polyphenols after RSA were established as follows: 53% methanol(V/V)as solvent, ratio of solvent to material 12:1(V/W), microwave power 510W and extraction 90s with the yield of total tobacco polyphenols 27.21mg/g. Tobacco polyphenols were identified by HPLC-PDA-MS, The main component in tobacco polyphenols were chlorogenic acid and rutin.
