板蓝根及其制剂中精氨酸和脯氨酸的配位毛细管电泳检测

摘    要:以配位作用毛细管电泳为手段,采用非衍生直接紫外吸收检测,对板蓝根注射液中各组分进行分析.考察运行缓冲液的浓度和酸度、运行电压、进样时间以及毛细管内径等因素对分离和检测的影响.在优化条件下,即运行缓冲液为30 mmol/L CuSO_4水溶液,pH=3.26,分离电压22 kV,位差进样40 S,毛细管内径50.0μm,检测波长254 nm,对其中含量最多的2种氨基酸--精氨酸和脯氨酸进行定量分析.分别考察其线性范围、检测限以及重现性.结果测得板蓝根注射液中精氨酸的含量为0.63 mmol/L,脯氨酸的含量为25.7 mmol/L.并在相同的优化条件下,对天然产物板蓝根提取物进行分析.结果测得其中精氨酸的质量分数为0.027,脯氨酸的质量分数为0.054.
