双酚A对小鼠肠道黏膜结构影响的观察

摘    要:为检测双酚A(BisphenolA)对小鼠小肠结构的影响,选用50只30g左右雌雄不拘,但同品系的昆明小白鼠作为试验对象。将小鼠随机分为5组,分别为双酚400、300、225mg/kg体重组、空白对照组和溶剂对照组。各组按0.1mL/100g体重灌喂小鼠。于给药后第6天全部处死,取十二指肠和空肠进行固定,用H.E.染色观察肠黏膜结构,并检测肠黏膜绒毛长度和宽度。同时,计数上皮内杯状细胞(GC)的数量变化;用Gomori氏改良法检测肠黏膜碱性磷酸酶(AKP)活性的变化。结果表明:各试验组小鼠空肠绒毛长度和宽度明显低于对照组,且差异显著(P〈0.05);各试验组小鼠十二指肠和空肠黏膜上皮内杯状细胞(GC)的数量均显著低于对照组(P〈0.05或P〈0.01);各试验组小鼠十二指肠和空肠黏膜单位面积内AKP阳性物所占面积的百分比均显著低于对照组(P〈0.05或P〈0.01)。由此推断双酚A对小鼠小肠黏膜结构有一定的损害作用,可能会影响其消化吸收功能。

Abstract:The effect of bisphenol A on the structure of small intestine is investigated. About 30 g Kunming mice with the same gender female or male were randomly divided into five groups, ten for each:bisphenol A 400 mg/kg,300 mg/kg,225 mg/kg, the control group and the solvent group. The four group were given orally with the dose of 0.1 mL/100g wb. The experiment lasted six days,then mice were sacrificed, their duodenums and jejunum were chose to observe the changes of the height and the width of villus of gut mucosa by the way of H E staining. Besides, the activity of AKP was tested through the Gomori staining.The statistics showed that the height and the width of villus of jejunum in the tested groups were decreased compared with the control group, the numbers of goblet cell in gut mucosa in the expeiment groups also decreased evidently and the activity of AKP in the experimental groups turned out to be remarkably decreased (p<0.05 or p<0.01). Accordingly, it can be concluded that the bisphenol A can hurt the structure of intestine mucosa, effect the absorb-ability.
