摘    要:文中对微生物学实验教材上介绍的Schaeffer-Fulton氏芽孢染色法进行了改进。用石碳酸复红试剂代替0.5番红水溶液作为复染剂,并对比不同石碳酸复红浓度及染色时间对染色效果的影响。结果表明,以石碳酸复红的10倍稀释液为复染剂,复染30 s~1 min时能达到最佳的染色效果。改进方法操作简单易行,染色效果对比鲜明,可使学生更易将菌体与芽孢区分开来。将改进的染色方法运用于微生物学实验教学,收到了良好的教学效果。

Abstract:Schaeffer- Fulton's spore staining method introduced in the textbooks of experimental microbiology was optimized in our study. 0. 5% safranine solution dye was replaced by carbolfuchsin dye, and the staining effects were explored in terms of carbolfuchsin concentrations and staining time. The results showed that the best staining results could be achieved with 1/10 carbolfuchsin dye, staining 30 seconds to 1 minute. The improved method was easy to operate, the staining effect was obvious, and the structure of mycelium and spore was easier to be distinguished. This method has been put into microbiological experiment teaching, and achieved better teaching results.
